He was born to Nandamuri Harikrishna and Shalini Bhaskar Rao on May 20, 1983, in Hyderabad, Telangana, India. His grandfather, N. T. Rama Rao Jr. is the son of Nandamuri Harikrishna (a Telugu actor and politician) and Shalini Bhaskar Rao (also a Telugu actor and politician). A grandson of N. T. Rama Rao, a famous Telugu actor and a former chief minister of Andhra Pradesh, he is an actor in his own right. He attended both Vidyaranya High School and St. Mary’s College, also located in Hyderabad, for his formal education. He has studied Kuchipudi dance and is proficient in the style.

N. Chandrababu Naidu, the current chief minister of Andhra Pradesh, is a nephew of Nandamuri Balakrishna and a cousin of Taraka Ratna and also of politician Nara Lokesh, making Jr. NTR the half-brother of actor and producer Nandamuri Kalyan Ram.

Rama Rao made his acting debut in his grandfather N. Rama Rao’s film Brahmarshi Vishwamitra (1991). A. T. R. Rama Rao. After that, director K. Raghavendra Rao recommended Rama Rao to S.
