Janani was born in the Chennai neighbourhood of Kathivakkam. She was accepted to Wollongong University in Australia after completing her undergraduate degree. She abandoned her academic plans to join the cast of Avan Ivan instead.

She tried out modelling after finishing her degree and ended up being in more than 150 TV commercials in her home state alone. Janani, on the other hand, insisted that acting was her “passion in life” and that she had always hoped to pursue a career in the field. As a model, she made a cameo appearance in Thiru Thiru Thuru Thuru (2009), and she worked as an assistant director alongside K. S. Ravikumar in Gautham Menon’s Vinnaithaandi Varuvaayaa (2011). Janani auditioned for director Bala, who was looking for a Tamil-speaking novice for the lead female character in his film project, Avan Ivan, in the early 2010s on the recommendation of director A. L. Vijay (2011). Bala signed her for the role after watching her audition, claiming that she “fits into the role.” Iyer’s acting as an innocent police policeman opposite Vishal in the comedy-drama film was praised by critics. Her subsequent film, Paagan (2013), starred her opposite Srikanth. Sify’s critic gushed, “Janani Iyer looks gorgeous and it is a relief to watch a Tamil girl speaking in her own voice.” Both her Bollywood debut and Malayalam debut movies, 3 Dots, were commercial flops.
She co-starred with Ashok Selvan in the 2014 criminal thriller Thegidi, which was met with critical acclaim and ended up being an unexpected financial success. “Janani Iyer is charming and performs her part brilliantly as Madhu the naïve girl,” wrote one critic for IndiaGlitz.com. In 2018, she made an appearance on Bigg Boss Tamil 2, a reality show presented by Kamal Haasan and broadcast on Star Vijay.
